**Experimental Principle:** The assay is based on the double-antibody sandwich enzyme-linked immunosorbent assay (ELISA) technique. A microtiter plate is pre-coated with a specific monoclonal antibody against human G6PD. After adding the sample, G6PD binds to the immobilized antibody. An HRP-conjugated secondary antibody is then added, forming an immune complex. Following thorough washing, the substrate TMB is introduced. Under the catalytic action of HRP, TMB turns blue and is then converted to yellow by the addition of a stop solution. The intensity of the color is directly proportional to the concentration of G6PD in the sample. The absorbance is measured at 450 nm using a microplate reader, and the G6PD concentration is calculated from a standard curve.
